EZ-Tn5™ Custom Transposome Construction Kit New

The INCOGNITO™ RNA Transcription Kits recently introduced by CELLSCRIPT™ is unique in what it brings for the process of in vitro transcription. The kit contains modified nucleosides, such as pseudouridine (Ψ) and/or 5-methylcytidine (m5C) in place of the corresponding U or C canonical nucleosides (Figure 1).

These mRNAs are also called "INCOGNITO mRNAs" since they do not initiate any innate immune responses like their analogous unchanged counterpart upon transfection into mammalian cells that express a wide range of RNA sensors.

It has been demonstrated that INCOGNITO mRNA, besides being less immunogenic, is translated into protein at much higher levels than the corresponding mRNA that does not contain modified nucleosides, both in cultured cells (Figure 2) and in whole organisms (Figure 3).

INCOGNITO mRNA have potential for therapeutic use in regenerative medicine such as tissue and organ repair, cell reprogramming, tissue engineering etc. by compensating the missing or defective protein once delivered to the cells in culture or in vivo.
